Opening minutes of season premiere:

Sydney delivers some major butt kicking to her former lover. JG says "I was strangling him. I had this electric cord around his face, and it was in his mouth at one point". MV says "There's a part where she turns around and kicks me in the stomach." MV then goes on to say "Jesus, her legs are like pistons. I was wearing a protective shell, but still--that girl gets props for her impact skills."

Melissa George's character (aka Mrs. Vaughn) is no longer recurring. She appears in ep 2.

Francie and Will are both MIA in the first episode.

Jack has left the CIA.

Sloane is living in Zurich, professing to be a good guy.

The Covenant seems to play a role in Syd's disappearance.

Weiss and Syd are working together and are roomies (I assume he's her new roommate).

Marshall is becoming a father.

Dixon is head of CIA.

And finally Justin Theroux (Syd's new love interest) will appear in episode 4 as a mysterious gun-for-hire who apparently hit the sheets with Syd during those missing 2 years.
